Uh well, if you are looking for a way to limit access to an area then you can do so by manipulating a trap chest. I built one under Neuro's realm for example that will only allow you to enter if you have perms on the realm. Basically it works because only people added to the realm can open the trap chest which sends a RS signal to open the door. So I suppose a design like this could be modified to somehow involve the manipulation of items.
In terms of actually making it so you need a valid key or something to open a door without giving people perms on the realm, thats a lot more tricky. Pretty much the problem is that you can't really make a custom object that anyone else couldn't make. So for example, if you were thinking of making a device where you need to drop a written book (which would serve as like an ID or invitation or whatever I guess) into a hopper to open a door, I can't think of a way to make it discernible from every other written book any old person could make.
One other idea maybe could be like a safe. So basically it would be an item sorter that would only allow you to enter a door if you drop the correct items in a hopper in the correct order. So like if you drop a piece of blue wool, a piece of gravel and one diamond in a hopper in that order, you can enter a door. But any RS doing something like this though is above me.
Lol I'd say your best chance if you want to limit access or something by the use of an "ID card" would be to have a bouncer at the door who would turn people away for not having a proper invitation hehe.
